In 2005, she made her first theatrical film appearance as Sunita in the musical romantic drama film Parineeta. It stars Saif Ali Khan, Sanjay Dutt, and Vidya Balan.

Supriya was cast as Urmi (Fuppu) in the 2010 Hindi family comedy-drama film Do Dooni Chaar. The film won the National Film Award for “Best Feature Film in Hindi” at the 58th National Film Awards.

She played the role of Seenu’s mother in the 2014 Hindi-language romantic action comedy film Main Tera Hero, starring Nargis Fakhri, Varun Dhawan, and Ileana D’Cruz.
